Abstract
AIM OF THE STUDY:
Total flavonids of Polygonatum(P) odoratum (TFP) were tested for anti-diabetic activity in streptozotocin (STZ)-induced diabetic mice and alloxan-induced diabetic rats.
MATERIALS AND METHODS:
Rhizoma Polygonati Odorati, well-known Chinese traditional medicine, is widely used for treatment of diverse diseases for example diabetes. In our study, TFP was extracted by 70% ethanol and purified by macroreticular resin. The experiments were designed to detect the anti-diabetic activity of TFP by determination of blood glucose (BG) using one touch gluco-meter and insul. levels by using a radioimmunoassay kit in streptozotocin (STZ)-induced diabetic mice and alloxan-induced diabetic rats and alpha-amylase inhibitory activity by alpha-amylase inhibition assay in vitro.
RESULTS:
TFP had beneficial effects on regulation of blood glucose. Daily administration with 50-200 mg/kg body weight of TFP for 9 days can reduce significantly hyperglycemia in STZ-induced diabetic mice. Thirtieth day administration with TFP (50-200 mg/kg body weight) also decreased significantly fasting blood glucose in alloxan-induced diabetic rats. The hypoglycemic effect of TFP at 50 and 100 mg/kg is less than that of acarbose 20 mg/kg and gliclazide 15 mg/kg. The hypoglycemic effects of TFP at 200 mg/kg is similar to that of acarbose 20 mg/kg and gliclazide 15 mg/kg. TFP also could increase significantly the insul. level in alloxan-induced type 2 diabetic rats (P<0.05) compared with control. Alpha-amylase inhibition assay in vitro showed that TFP inhibited significantly alpha-amylase activity in a dose-dependent manner.
CONCLUSIONS:
TFP possess significant dose-dependent anti-diabetic activity. TFP is one of the primary hypoglycemic active compounds of Polygonatum odoratum which would worth further study and development.

Abstract
Rhizoma Polygonati falcatum (RPF) has been used as a traditional herbal medicine in Asia, because of its anti-hyperglycemic, anti-triglycemic, and anti-tumor activity. In this study, we determined the anti-adipogenic potential of RPF extract and its component kaempferol in 3T3-L1 adipocytes, and the underlying molecular mechanism(s) using microarray analysis. Adipocyte differentiation of 3T3-L1 cells was significantly impaired by RPF extract and kaempferol as monitored by Oil Red O staining and quantitative measurement of lipid accumulation. Additionally, the mRNA expression of adipogenesis genes decreased on treatment with kaempferol. The role of kaempferol at the genome-wide level was further assessed by a microarray approach. Our analysis indicated that kaempferol decreased the expression of adipogenic transcription factors (Pparγ, Cebpβ, Srebp1, Rxrβ, Lxrβ, Rorα) and genes involved in triglyceride biosynthesis (Gpd1, Agpat2, Dgat2), while increasing lipolysis-related genes, such as Tnfα, Lsr, and Cel. Finally, co-transfection assays using luciferase reporter gene and reverse transcription-polymerase chain reaction (RT-PCR) analysis using peroxisome proliferator-activated receptor-γ (PPARγ) target genes indicated that kaempferol significantly repressed rosiglitazone-induced PPARγ transcriptional activity. Overall, our data suggests that kaempferol, a major component of RPF, may be beneficial in obesity, by reducing adipogenesis and balancing lipid homeostasis partly through the down-regulation of PPARγ.

Abstract
ETHNOPHARMACOLOGICAL RELEVANCE:
Rhizoma Polygonati is originated from the dried rhizomes of Polygonatum sibircum Red. It has long been used in traditional Chinese medicine for the treatment of inflammatory disorders.
AIM OF THE STUDY:
The present study aims to investigate the anti-inflammatory effect of aqueous extract of Rhizoma Polygonati (ERP) in a mouse model of inflammation induced by 12-O-tetradecanoylphorbol-acetate (TPA).
MATERIALS AND METHODS:
The anti-inflammatory effect was evaluated by measuring the ear thickness and activity of myeloperoxidase (MPO). The anti-inflammatory mechanism was explored by determining the protein and mRNA levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, and IL-6.
RESULTS:
The results showed that ERP significantly decreased the ear thickness and MPO activity in mouse model of inflammation induced by TPA. In addition, ERP also remarkably inhibited the protein and mRNA levels of iNOS, COX-2, TNF-α, IL-1β, and IL-6.
CONCLUSIONS:
These results indicate that ERP has potential anti-inflammatory effect on TPA-induced inflammatory in mice, and the anti-inflammatory effect may be mediated, at least in part, by inhibiting the mRNA expression of a panel of inflammatory mediators including iNOS, COX-2, TNF-α, IL-1β, and IL-6.
